Effects of peroxidizing herbicides on protoporphyrin IX levels in non-chlorophyllous soybean cell culture

Abstract The mode of action of 16 peroxidizing herbicides belonging to four different families (diphenyl ethers, oxadiazon, pyridine derivatives, and pyrazole derivatives) has been studied in nonchlorophyllous soybean cell cultures. Whenever possible, we have compared active and inactive compounds. Phytotoxic effects were estimated on the basis of growth inhibition, either in the dark or in the light. Protoporphyrin IX accumulations were estimated in dark-treated samples, using a simple methodology. Characterization of (3H) acifluorfen binding to purified pea etioplasts, and evidence that protoporphyrinogen oxidase specifically binds acifluorfen

It is now generally accepted that protoporphyrinogen oxidase is the target-enzyme for diphenylether-type herbicides. Recent studies [Camadro, J-M., Matringe, M., Scalla, R. & Labbe, P. (1991) Biochem. J. 277, 17–21] have revealed that in maize, diphenyl ethers competitively inhibit protoporphyrinogen oxidase with respect to its substrate, protoporphyrinogen IX. In this study, we show that, in purified pea etioplast, [3H]acifluorfen specifically binds to a single class of high-affinity binding sites with an apparent dissociation constant of 6.2 ± 1.3 nM and a maximum density of 29 ± 5 nmol/g protein. Photodynamic therapy of nodular basal cell carcinoma with multifiber contact light delivery.

To overcome the limited treatment depth of superficial photodynamic therapy we investigate interstitial light delivery. In the present work the treatment light was delivered using a system in which three or six clear-cut fibers were placed in direct contact with the tumor area. This placement was thought to represent a step toward general purpose interstitial PDT. Twelve nodular basal cell carcinomas were treated employing delta-aminolevulinic acid and 635 nm laser irradiation. Hole burning and pressure phenomena in chromoproteins

Abstract We investigated the behavior of spectral holes under pressure at various frequencies within the inhomogeneous band for two proteins, namely myoglobin and horseradish peroxidase. In order to achieve narrow bandwidth hole burning, the heme chromophore was replaced by protoporphyrin IX and mesoporphyrin IX, respectively. In myoglobin, we found that the pressure induced shift of the holes varied in a strongly non-linear fashion, when the burn-frequency was tuned across the absorption band. In horseradish peroxidase the pressure shift was linear with burn-frequency but changed in a dramatic fashion upon complex formation with a substrate molecule. Thermal broadening of the Soret band in heme complexes and in heme-proteins: role of iron dynamics

We report the thermal broadening of the Soret band in heme-CO, heme-OH and protoporphyrin IX in the temperature range 300-20 K. For protoporphyrin IX the temperature dependent Gaussian line broadening follows the behavior predicted by the harmonic approximation in the entire temperature range investigated. In contrast, for heme-CO and heme-OH the harmonic behavior is obeyed only up to about 180 K and an anomalous line broadening increase is observed at higher temperatures. This effect is attributed to the onset of anharmonic motions of the iron atom with respect to the porphyrin plane. Abstract Age-related changes in joint tissues lead to osteoarthritis (OA). Detection of early changes in OA patients may help to initiate treatments before the establishment of irreversible joint destruction. STR/ort mice develop with age a severe degenerative joint disease that resembles human OA thus allowing the investigation of biochemical markers as well as new treatments in an accelerated time frame. Cellular damage to human hepatocytes through repeated application of 5-aminolevulinic acid.

Abstract Background/Aims : 5-Aminolevulinic acid (ALA), a precursor of porphyrins is used for photodynamic diagnosis and therapy within topical or systemic applications. A potential toxic effect on the human liver is of major interest and therefore we investigated the impact of a repeated application of ALA without illumination on cultures of human hepatocytes. Methods : After ALA treatment of hepatocytes in vitro the porphyrin synthesis, albumin secretion, liver-specific enzyme release, and malondialdehyde levels were determined. In vitro and in vivo porphyrin accumulation by C6 glioma cells after exposure to 5-aminolevulinic acid

Several malignant tissues synthesize endogenous porphyrins after exposure to 5-aminolevulinic acid (5-ALA). The present experiments have been designed to elucidate whether the C6 glioma cell, a model cell for human malignant glioma, similarly synthesizes porphyrins when exposed to 5-ALA, and whether specific synthesis occurs when C6 cells are inoculated into rat brains to form a tumor. In this situation the blood-brain barrier may interfere with 5-ALA availability, and spreading of porphyrins with edema outside the tumor may occur. Highly soluble multifunctional MnO nanoparticles for simultaneous optical and MRI imaging and cancer treatment using photodynamic therapy

Superparamagnetic MnO nanoparticles were functionalized using a hydrophilic ligand containing protoporphyrin IX as photosensitizer. By virtue of their magnetic properties these nanoparticles may serve as contrast enhancing agents for magnetic resonance imaging (MRI), while the fluorescent target ligand protoporphyrin IX allows simultaneous tumor detection and treatment by photodynamic therapy (PDT). Caki-1 cells were incubated with these nanoparticles. Subsequent exposure to UV light lead to cell apoptosis due to photoactivation of the photosensitizer conjugated to the nanoparticles. High-grade gliomas (HGGs) are the most frequent primary malignant brain tumors in adults, which lead to death within two years of diagnosis.
